Dual role of prostaglandin D2 in the regulation of Eescherichia coli-induced endometrial inflammatory response in dairy cows
摘要
Abstract
This study utilizes an in vitro cultured bovine endometrial tissue model to elucidate the mechanism of prostaglandin D2(PGD2)in the pathogenesis of Escherichia coli-induced endometritis.By analyzing different infection time,this study aims to investigate the role of endogenous PGD2 in regu-lating uterine endometrial tissue damage and the expression of related molecules during E.coli infection.Additionally,it further explores the regulatory effect of PGD2 on the secretion of inflammatory cytokines(IL-1β,IL-10,IL-8),thereby revealing the potential mechanisms of PGD2 in the occurrence and progression of bovine endometritis.The results showed that the PGD2 synthase inhibitors(H-PGDS inhibitor 1 and HQL-79)significantly inhibited the secretion of PGD2 in bovine endometrial tissues at 9 and 24 h post-E.coli infection(P<0.000 1).At 9 h post-E.coli infection,inhibition of PGD2 synthesis significantly alleviated the damage caused by E.coli to bovine endometrial tissues and notably reduced the expression of the damage-associated factor HABP-2(P<0.01).At the same time,inhibition of PGD2 synthesis reduced the secretion of IL-1β and IL-8,while significantly increasing the secretion of IL-10.However,at 24 h post-E.coli infection,a completely opposite result was observed,where the inhibition of PGD2 synthesis led to a further exacerbation of the inflammatory response to some extent.These results indicate that the effects of endogenous PGD2 on bovine endometrial tissue during E.coli infection exhibit a clear time-dependent manner,with its immunoregulatory functions showing phase-specific variations throughout the course of infection.关键词
