Effect of BLP on cytokine secretion of bovine bone marrow-derived macrophages induced by E.coli infection

Abstract
To investigate the mechanism of action of Braun lipoprotein(BLP)in Escherichia coli-induced bovine endometritis,this study stimulated macrophages with LPS,BLP,and LPS combined with BLP;and infected bovine bone marrow-derived macrophages(bBMMs)with E.coli JE5505,E.coli DH5α,and E.coli JE5505 combined with BLP stimulation.ELISA,quantitative real-time PCR,and Western-blot techniques were employed to analyze cytokine secretion,the activation of inflammatory and anti-inflammatory signaling pathways,and the effect of TLR2 receptor activation on cytokine production.Compared with E.coli JE5505 infection group,bBMMs infected with E.coli DH5α group or infected with E.coli JE5505 combined with BLP stimulation group exhibited significantly decreased levels of pro-inflammatory cytokines TNF-α and IL-1β,while the secretion of the anti-inflammatory cytokine IL-10 was significantly elevated.Western-blot analysis demonstrated that BLP modulated the activation of the NF-κB/MAPK pathway as well as the expression of the anti-inflammatory signaling pathway STAT3 in bBMMs.These findings suggest that during E.coli infection,BLP is recognized by the TLR2 receptor and may attenuate the production of pro-in-flammatory cytokines and chemokines in bBMMs through the activation of the MAPK and NF-κB signaling pathways.Additionally,BLP may enhance the production of the anti-inflammatory cytokine IL-10 via the activation of the STAT3 signaling pathway,thereby mitigating the inflammatory response induced by E.coli infection.This indicates that BLP has potential therapeutic effects in the regulation of infection-induced inflammation.关键词
