Analysis of Tick Species and Genetic Diversity of Rhipicephalus turanicus in Border Areas of Xinjiang

Abstract
To investigate tick species composition and the genetic diversity of Rhipicephalus turanicus in Qapqal Xibe Autonomous County and Huocheng County,Xinjiang Uygur Autonomous Region,morphological identification was used to preliminarily classify tick species.The mitochondrial 16S rRNA gene sequences were amplified and sequenced for genetic analysis.A phylogenetic tree was constructed using MEGA 11.0 software,and reverse transcription-quantitative polymerase chain reaction(RT-qPCR)was used to detect the presence of severe fever with thrombocytopenia syndrome virus(SFTSV),tick-borne encephalitis virus(TBEV),Alongshan virus(ALSV),and Crimean-Congo hemorrhagic fever virus(CCHFV).Genetic diversity analysis was performed for the dominant tick species.Among the 670 collected ticks,three genera and species were identified,with Rhipicephalus turanicus being the dominant species(67.16%,450/670),followed by Hyalomma asiaticum(31.79%,213/670)and Dermacentor marginatus(1.04%,7/670).There was a significant difference in species distribution between the two counties(χ²=19.733,P<0.01).All tick samples tested negative for the four viruses.Analysis of 24 Rhipicephalus turanicus 16S rRNA gene sequences revealed a nucleotide diversity(Pi)of 0.010 65 and a haplotype diversity(Hd)of 0.873,with 13 haplotypes identified.Neutrality tests and mismatch distribution indicated no recent population expansion in the four populations.Analysis of molecular variance(AMOVA)showed that 94.52%of genetic variation originated within populations,with only 5.48%among populations.These findings suggest that Rhipicephalus turanicus is the dominant species in the surveyed regions and exhibits high genetic diversity,highlighting its adaptability to various environments and contributing valuable data for understanding tick biodiversity and tick-borne diseases.关键词
