藏猪源2型猪链球菌全基因组序列分析
Whole-genome Sequence Analysis of Streptococcus suis Serotype 2 Isolated from Tibetan Pigs

Abstract
To elucidate the genomic characteristics and evolutionary relationship of Streptococcus suis serotype 2 derived from Tibetan pigs,we performed whole-genome sequencing of strain LD—isolated from the lungs of a diseased Tibetan pig—using Illumina HiSeq second-generation and Nanopore third-generation sequencing technologies.Bioinformatics methods were employed for genome annotation and phylogenetic analysis.The complete genome of strain LD was 2 714 470 bp in length,comprising 2 521 genes.The proportions of annotated genes in the COG,GO,KEGG,CAZy,and PHI-base databases were 80.00%,28.00%,56.29%,2.54%,and 17.49%,respectively.Virulence factor analysis identified 132 virulence genes,among which 37.87%were involved in evading the host immune system.Antibiotic resistance analysis revealed 83 resistance genes,mainly related to four major classes of antibiotics,predominantly macrolides.Protein sequence analysis predicted 78 signal peptide-containing proteins.A total of 378 genes associated with transport proteins were identified using the Transporter Classification Database(TCDB),and 569 transmembrane proteins were predicted via the TMHMM tool.Phylogenetic analysis showed that strain LD was most closely related to strains U(GCF_002964045.1)isolated from Hubei,China in 2018 and W(GCF_902702775.1)isolated from the Netherlands in 2019.This study represents the first genome-wide analysis of S.suis serotype 2 isolated from Tibetan pigs,enriching the genomic dataset of S.suis and providing valuable data support for the health management and disease prevention of Tibetan pigs in Ganzi Prefecture.关键词
