皂角刺总皂苷的提取工艺、化学成分和体外抗炎活性研究
Study on the Extraction Process,Chemical Composition and in vitro Anti-inflammatory Activity of Total Saponins from Gleditsia sinensis Lam

Abstract
To explore the extraction process,chemical composition,and in vitro anti-inflammatory activity of total saponins from Gleditsia sinensis Lam.(GTS),and to provide a reference for the development of novel traditional Chinese veterinary additives,this study employed single-factor experiments and response surface methodology to optimize the extraction process of GTS.The chemical components of GTS were preliminarily identified using ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-QTOF-MS).Furthermore,the effects of GTS on cell viability,nitric oxide(NO)production,and phagocytic activity were evaluated using a lipopolysaccharide(LPS)-induced inflammation model in RAW 264.7 mouse macrophages.The optimal extraction conditions were determined as follows:ethanol concentration 44%,liquid-to-material ratio 50∶1(mL/g),extraction time 20 minutes,extraction temperature 40℃,and two extraction cycles,resulting in an average yield of(1.44±0.01)%.The major constituents of GTS were preliminarily identified as 13 saponins and 3 flavonoids.GTS showed no cytotoxicity to RAW 264.7 cells and significantly inhibited LPS-induced increases in NO production and phagocytic activity(P<0.01),with no significant difference compared to control drug dexamethasone(DEX)(P>0.05).These findings suggest that GTS possesses notable anti-inflammatory potential and may serve as a valuable candidate for the development of new Chinese veterinary medicine additives.关键词
