Optimizing the fermentation medium of biocontrol Streptomyces PBSH9 against potato common scab using response surface methodology

Abstract
Potato common scab is a significant disease that restricts the development of the potato industry.This study screened the biocontrol strain PBSH9,and its metabolites showed significant inhibitory effects on the pathogen,achieving a field control efficacy of 73.97%.To identify the active compounds,the secondary metabolites of strain PBSH9 were iso-lated and purified,successfully yielding the active substance.To enhance the yield of this active substance,response sur-face methodology(RSM)was employed to optimize its culture medium.Based on single-factor experiments and Plack-ett-Burman tests,combined with the steepest ascent method and Box-Behnken design response surface methodology to fit the nonlinear equation between significant factors and the yield of active compound,the optimal fermentation medium for-mula was obtained.The optimized formula was 4.50 g·L-1 glucose,50.00 g·L-1 soluble starch,15.00 g·L-1 corn flour,21.08 g·L-1 soybean meal,2.00 g·L-1 peptone,0.50 g·L-1(NH4)2SO4,0.60 g·L-1 MgSO4,and 0.04 g·L-1 KH2PO4.The mod-el-predicted yield of active compounds was 199.92 μg·mL1,while the actual value was 184.35 μg·mL-1,which was 5.83 times higher than the yield of compounds in the basic culture medium.This study provides crucial technical sup-port for the development of efficient biopesticide products against potato common scab.关键词
