基于血清代谢组学探讨逍遥散加减方对肉鸡肺动脉高压综合征的影响
Mechanism of modified Xiaoyao San prescription on pulmonary hypertension syndrome in broilers based on serum metabolomic analysis

Abstract
In this experiment,non-targeted metabolomics was used to explore the effect of modified Xiaoyao San prescription on serum metabolites of pulmonary hypertension syndrome(PHS)in broilers and scientifically evaluate its therapeutic effect.The formula contains 10 herbal medicines which are Radix Bupleuri,Angelica Sinensis,Paeoniae Radix Alba,Rhizoma Atractylodis Macrocephalae,Poria,Radix Glycyrrhizae,Hebra Menthae,Radix Salviae Miltiorrhizae,Stigma Maydis,and Rabdosiae Serrae.The above ingredients were formulated in a mass ratio of 3∶4∶3∶3∶3∶3∶2∶3∶3∶3.The experimental chickens were divided into three groups:control group,PHS group,and Xiaoyao San treatment group.At 20 days of age,broilers in the control group were injected with normal saline through the wing vein,and those in the PHS group and Xiaoyao San treatment group were injected with CM-32 cellulose suspension to induce PHS.In the meanwhile,the broilers in treatment group were fed with herbal compound in feed.The total PHS incidence in each group was counted at the end of the experiment.At 42 days of age,serum was collected from broiler chickens for LC-MS/MS metabolomics analysis,and heart samples were collected to calculate scites heart index(right ventricular weight/total ventricular weight).The results showed that PHS morbidity in PHS group was significantly higher than that in the control group(P<0.05),while the PHS morbidity in Xiaoyao San treatment group was significantly lower than that in the PHS group(P<0.05).Compared with the control group,the ascites heart index(AHI)of the PHS group was extremely significantly increased(P<0.01),while the AHI of Xiaoyao San treatment group was extremely significantly decreased compared with the PHS group(P<0.01).A total of 64 metabolites with a significant difference were screened out for the PHS vs control group,among which 27 metabolites were up-regulated and 37 metabolites were down-regulated.A total of 26 metabolites with a significant difference were screened out for Xiaoyao San treatment group vs PHS group,among which 7 metabolites were up-regulated and 19 metabolites were down-regulated.Nine metabolites with significant differences were identified within the three groups.Two metabolites showed regularly fluctuations,which were 6-hydroxymelatonin and N-acetylglutamine.It is speculated that these two metabolites are involved in the pathogenesis and treatment of PHS through their anti-oxidative stress effects.Xiaoyao San can play a therapeutic effect on PHS by regulating the expression of N-acetylglutamine and 6-hydroxymelatonin.The newly discovered metabolites are expected to be further applied in the study of the pathogenesis and prevention of PHS.关键词
