Extraction optimization and content determination of total flavonoids from Operculina turpethum(Linn.)S.Manso originating from different geographical sources

Abstract
The objective was to optimize the extraction protocol for total flavonoids from Operculina turpethum(Linn.)S.Manso,establish a reliable method for its content determination,and provide a robust technical basis for subsequent investigations of this species.Extraction yield of total flavonoids was employed as the sole evaluation criterion.Single-factor experiments were conducted to investigate the individual effects of ethanol concentration,liquid-solid ratio,and ultrasonic duration on the yield.On the basis of these results,L9(34)orthogonal array design was adopted to finalize the optimal conditions.The validated protocol was subsequently applied to quantify total flavonoids in O.turpethum samples collected from different geographical origins.The results showed that the optimal parameters were identified as 70%ethanol,a liquid-solid ratio of 15∶1(mL∶g),and an ultrasonic treatment time of 40 min.Under these conditions,the total flavonoid content in samples of varying provenance ranged from 0.68%to 1.79%,expressed as rutin equivalents.The optimized procedure demonstrates excellent repeatability and stability,thus providing a reliable method for routine extraction and quantification of total flavonoids from O.turpethum.Notably,flavonoid levels differ significantly among samples from different geographical origins,highlighting the importance of provenance-based quality control.关键词
