Optimization of extraction technology of catechin and phloridzin in Cynomorii Herba by Box-Behnken response surface methodology

摘要

Abstract

The method was established for the content determination of catechin and phloridzin by HPLC.Based on the single-factor experiments of methanol concentration,liquid-solid ratio,extraction time,extraction temperature,ultrasonic power,and extraction times,three main influencing factors namely methanol concentration,liquid-solid ratio,and extraction time were selected to optimize the extraction process parameters of catechin and phloridzin from Cynomorii Herba with Box-Behnken experimental design and response surface analysis method.The results showed that catechin had a good linear relationship in the range of 1.64-24.60µg,and phloridzin in the range of 0.04-0.60µg(R2>0.999 0),and the specificity,precision,repeatableility,stableility were all good.The methanol concentration,liquid-solid ratio,and extraction time were highly significantly correlated with the extraction amounts of catechin and arbutin.The optimal extraction process was as follows:extraction solvent 90%methanol,liquid-solid ratio 30∶2(mL∶g),extraction temperature 30℃,extraction time 30 min,and extraction times 2.The established method of extraction and content determination with catechin and phloridzin from Cynomorii Herba in this study are effective and feasible,providing a theoretical basis and technical support for the quality evaluation and the development and application of catechin and phloridzin in Cynomorii Herba.
