摘要

Abstract

Objective To investigate the lipid-lowering effects and mechanisms of Jianpi Qushi Formula(JPQSF)in high-fat diet-induced obese mice.Methods Male C57BL/6J mice were randomly divided into normal group(n=6)and high-fat diet(HFD)model group(n=44).The normal group received standard maintenance chow,while the HFD group was fed a high-fat diet for 12 weeks.Obesity modeling was considered successful when body mass of HFD mice exceeded the average mass of normal mice by 20%.Thirty successfully modeled obese mice were randomly assigned to model group,orlistat group(0.046 8 g·kg-1),and low-,medium-,and high-dose JPQSF groups(6,12,and 24 g·kg-1),with 6 mice per group.All groups continued HFD feeding while receiving respective treatments via daily gavage for 8 weeks.Body mass was measured weekly at fixed time points.An oral glucose tolerance test(OGTT)was conducted 2 weeks before the end of the experiment,and the area under the time-blood glucose curve(AUC)was calculated.Epididymal white adipose tissue(eWAT),inguinal white adipose tissue(iWAT),and interscapular brown adipose tissue(iBAT)were dissected and weighed.Serum levels of total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)were measured.Pathological changes in adipose tissues were observed by hematoxylin-eosin(HE)staining.Real-time PCR was used to detect mRNA expression of UCP1,PRDM16,PGC-1α,and PPARγ in adipose tissues.Immunohistochemistry was employed to assess UCP1 protein expression.Results Compared with the normal group,the model group showed significantly increased body mass,adipose tissue mass(eWAT,iWAT,and iBAT),and AUC values(P<0.01);significantly elevated serum TC,TG,and LDL-C levels(P<0.05,P<0.01)and reduced HDL-C levels(P<0.01);markedly enlarged adipocyte volume and diameter with reduced cell number per unit area in eWAT and iBAT;significantly downregulated mRNA expression of UCP1,PGC-1α,PPARγ,and PRDM16 in eWAT and iBAT(P<0.01);and significantly decreased UCP1 protein expression in eWAT and iBAT(P<0.01).Compared with the model group,all treatment groups exhibited significantly reduced body mass from weeks 5 to 8(P<0.01).The low-and high-dose JPQSF groups showed decreased iWAT mass(P<0.05),while the medium-and high-dose JPQSF groups exhibited reduced eWAT and iBAT mass(P<0.05,P<0.01).All treatment groups had significantly lower AUC values(P<0.05,P<0.01).The high-dose JPQSF group showed significantly reduced serum TC,TG,and LDL-C levels(P<0.05,P<0.01)and increased HDL-C levels(P<0.05).All treatment groups demonstrated reduced adipose tissue volume,reduced adipocyte diameter,increased cell number per unit area,and denser arrangement in eWAT and iBAT.mRNA expression of UCP1,PGC-1α,and PPARγ in eWAT was significantly upregulated in all treatment groups(P<0.05,P<0.01).The medium-and high-dose JPQSF groups showed significantly increased UCP1,PGC-1α,and PPARγ mRNA expression in iBAT(P<0.05,P<0.01),while the high-dose group exhibited elevated PRDM16 mRNA expression in iBAT(P<0.01).UCP1 protein expression in eWAT and iBAT was significantly upregulated in all treatment groups(P<0.01).Conclusion JPQSF demonstrates significant anti-obesity effects in HFD-induced obese mice,effectively improving glucolipid metabolism disorders and pathological changes in adipose tissues,potentially mediated by promoting white adipose browning and brown adipose tissue activation.
