Abstract

Objective:To study the chemical composition changes of Tibetan medicine Berberidis Cortex and Phyllanthi Fructus before and after compatibility,and to explore the rationality of the compatibility of the two drugs.Methods:UPLC-Q-Or-bitrap MS was used to analyze the chemical constituents of the single decoction and combined decoction of Berberidis Cortex and Phyllanthi Fructus.The chemical constituents were identified by comparing with the ion fragment information of the reference substance,relevant literature and database.Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to reveal the chemical constituents of the single decoction and combined decoction of the two drugs.Results:A total of 33 chemical constituents were identified including 12 alkaloids,11 polyphenols,6 organic acids,2 fla-vonoids and 2 sugars.Through PCA and OPLS-DA analysis,it was found that there was a significant difference between the single decoction and the combined decoction of the two drugs.A total of 10 different components were screened according to Variable Importance in Projection(VIP)>3 and t test.The relative contents of berberine,jatrorrhizine,palmatine,bufotenidine,corilagin,6-O-galloylglucose,quinic acid,oblongine and fructose were significantly increased in the combined decoction,while the relative content of trehalose was significantly decreased in the combined decoction.Conclusion:After the compatibility of Berberidis Cortex and Phyllanthi Fructus,the dissolution of various active components in the combined decoction could be promoted.These components may be the material basis for the synergistic treatment of diseases by the compatibility of Berberidis Cortex and Phyllanthi Fructus.
