Exploring the Mechanism of Dendrobium Mixture Regulating Autophagy via NF-κB Signaling Pathway to Alleviate Pain in Diabetic Neuropathic Pain Model Rats

Abstract
Objective:To observe the therapeutic effect of Dendrobium Mixture on a rat model of diabetic neuropathic pain(DNP)and explore its potential molecular mechanism in regulating spinal cord cell autophagy to alleviate pain in diabetic neuropathic rats.Methods:Fifty SPF-grade male SD rats were randomly divided into a normal group(10 rats)and a model establishment group(40 rats).The model establishment group was fed a high-fat and high-sugar diet for 12 weeks,followed by two intraperitoneal injections of streptozotocin [STZ,25 mg/(kg·m²),with a 3-day interval] to induce DNP.At the 14th week,successfully modeled rats were randomly divided into a model group,a chloroquine group,and a Dendrobium Mixture group,with 10 rats in each group.The model group and the blank group were administered normal saline by gavage,the Dendrobium Mixture group was administered Dendrobium Mixture by gavage,and the chloroquine group was administered chloroquine by gavage,all at the dose of 1 mL/100 g,for a total of 4 weeks respectively.Blood glucose and pain threshold were monitored;levels of HS-CRP,FBG,INS,inflammatory cytokines(IL-1β,IL-6,TNF-α),and blood lipids(TG,TC,HDL,LDL)in serum were measured;Western blot was used to analyze the protein expression of NF-κB,p-NF-κB,LC3,Beclin-1,and p62 in the spinal cord segment(L4-L5).Results:Compared with the model group,the Dendrobium Mixture group significantly increased the pain threshold,improved glucose metabolism,regulated lipid metabolism,reduced the levels of inflammatory cytokines(IL-1β,IL-6,TNF-α)and HS-CRP,downregulated LC3 and Beclin-1,upregulated p62,and decreased the p-NF-κB/NF-κB ratio(P<0.01).Conclusion:Dendrobium Mixture alleviates inflammatory responses and increases the pain threshold by reducing glucose and lipid metabolism levels and inhibiting the release of immune-inflammatory cytokines in DNP model rats.The mechanism of action may be related the ability of Dendrobium Mixture to suppress pathological autophagy in spinal cord cells and inhibit the activation of the NF-κB signaling pathway.关键词
