摘要

Abstract

Objective:To observe the effect of Qinbai Qingfei Concentrated Pill on RanBP2-mediated TRPV1 SUMOylation,thereby elucidating its molecular mechanism for inhibiting airway neurogenic inflammation.Methods:Human neuroblastoma cell line(SH-SY5Y)was selected,and a PIC cell model was established by combined induction with bradykinin and capsaicin.The model was intervened with Qinbai Qingfei Concentrated Pill medicated serum and a RanBP2 inhibitor.Morphological changes in SH-SY5Y cells were observed under an optical microscope.The contents of neuropeptides SP and CGRP were detected by ELISA.The expression of RanBP2 and TRPV1 proteins was detected by immunofluorescence staining.TRPV1 protein SUMOylation was detected by co-immunoprecipitation.Based on aforementioned model,SH-SY5Y was co-cultured with human primary airway smooth muscle cells(HTSMC)using a Transwell system.Morphology changes in HTSMC cells were observed under an optical microscope.Cell migration levels were detected by a cell scratch assay.The contents of IL-6 and IL-8 were detected by ELISA.Results:Compared with the model group,Qinbai Qingfei Concentrated Pill significantly inhibited the secretion of SP and CGRP in SH-SY5Y cells,and down-regulated the expression of RanBP2 protein and the SUMOylation of TRPV1 protein(P<0.01).It also significantly inhibited the migration rate of HTSMC and the secretion of IL-6 and IL-8(P<0.01).Conclusion:Qinbai Qingfei Concentrated Pill can inhibit RanBP2-regulated SUMOylation of TRPV1 protein,thereby achieving the effect of suppressing PIC airway neurogenic inflammation.
