酒制丝瓜络治疗类风湿性关节炎的效果及初步机制分析
Effect of Wine-Processed Luffa cylindrica on Rheumatoid Arthritis and the Preliminary Mechanism Analysis

Abstract
Objective:To systematically evaluate the anti-rheumatoid arthritis activity of wine-processed Luffa cylindrica,and to preliminarily analyze this potential molecular mechanism.Methods:Forty SD rats were randomly divided into a blank group(8 rats)and a modeling group(32 rats).An adjuvant-induced arthritis model was prepared by intradermal injection of 0.1 mL of Freund's complete adjuvant into the left hind foot pad.After successful modeling,the rats in the modeling group were randomly divided into a model group,a positive group(with dexamethasone 0.075 mg/kg),a crude product group(with crude Luffa cylindrica 2.8 g/kg),and a wine-processed group(with wine-processed Luffa cylindrica 2.8 g/kg),with 8 rats in each group.Starting from the 10th day of the experiment,the blank group and the model group were administered an equal volume of normal saline by gavage,and the administration groups were administered the corresponding drugs by gavage,for an intervention period of 14 days.Behavioral performances of rats in each group were observed during the experiment.The toe volume and arthritis index score were measured every 7 days to evaluate the effect of wine-processed Luffa cylindrica on rats with adjuvant-induced arthritis.After the rats were sacrificed,the spleen and thymus indices were calculated.IL-1β and TNF-α levels were detected by ELISA.The protein expression of TNF-α and ADAMTS-4 in rat articular cartilage tissue and the protein expression of p38 MAPK and MIF in synovial tissue were detected by Western blot.The mRNA expression of TNF-α,MIF,ADAMTS-4,and p38 MAPK in rat synovial tissue was detected by RT-qPCR.Results:Compared with the rats in model group,the mental state and activity level of rats in the positive group,crude product group,and wine-processed group were significantly improved.The positive group and the various Luffa cylindrica groups increased the inhibition rate of toe swelling to varying degrees(P<0.05,P<0.01),and significantly reduced the serum levels of IL-1β and TNF-α,as well as the mRNA and protein expression of TNF-α,MIF,ADAMTS-4,and p38 MAPK(P<0.05,P<0.01,P<0.001).Conclusion:Wine-processed Luffa cylindrica has a good therapeutic effect on the adjuvant-induced arthritis model,with improvement of toe redness and swelling,and its mechanism may be related to down-regulation the expression of TNF-α,MIF,ADAMTS-4,and p38 MAPK,thereby reducing the inflammatory response.关键词
