摘要

Abstract

[Objective]The 4CL gene plays an important role in plant response to abiotic stress.This study aimed to clarify the function of the Pp4CL1 gene from Peucedanum praeruptorum Dunn under salt stress,so as to provide a foundation for further research into the mechanisms underlying the response to abiotic stress.[Method]Using P.praeruptorum Dunn as test material,the Pp4CL1 gene was cloned and subjected to bioin-formatically analysis.Overexpressing strains of Arabidopsis thaliana were obtained by genetic transformation,and three high-expression positive strains were selected for the NaCl stress tolerance test.[Result]Bioinfor-matics analysis showed that the Pp4CL1 gene coded for 543 amino acids(aa),with a molecular formula of C2693H4274N684O789S26 and the molecular weight of 59 691.23 Da,indicating it is a stable protein.The protein contained two conserved structural domains,lack of transmembrane structure,and its secondary structure mainly consisted of α-folding and extended chain.Under 150 mmol·L-1 NaCl treatment,Pp4CL1-overex-pressing A.thaliana enhanced salt tolerance,and the lines showed higher activities of superoxide dismutase(SOD),peroxidase(POD),catalase(CAT)and ascorbate peroxidase(APX),along with significantly increased chlorophyll,proline and total glutathione contents.Additionally,malondialdehyde content was significantly lower than that in the wild type.[Conclusion]The Pp4CL1-overexpressing Arabidopsis lines demonstrated stronger resistance to salt stress,indicating that the Pp4CL1 gene confers salt tolerance capabilities.
