Abstract

[Objective]To facilitate the early identification of eggs,larvae,and pupae of the tobacco moth(Ephestia elutella)and the cigarette beetle(Lasioderma serricorne),this study employed DNA barcoding tech-nology to conduct molecular identification of different developmental stages of these two tobacco storage pests,aiming to provide early warning for the prevention and control of stored tobacco pests.[Method]DNA was ex-tracted from four life stages of the tobacco moth and the tobacco beetle,including eggs,larvae,pupae,and adults.Species-specific primers were designed for PCR amplification of the mitochondrial DNA Cytochrome oxidase I(COI)gene from each life stage of the two pests.Subsequently,sequencing,alignment,and analysis of the ampli-fied sequences were conducted.[Result]Specific bands of the same size were amplified from the four life stages of both pests,indicating that the COI gene barcode could be used to identify tobacco moth and the cigarette beetle at all life stages,and the accuracy of the identification results was not affected by the DNA samples from different life stages.[Conclusion]The precise identification of the tobacco moth and the cigarette beetle can be achieved within a short time by amplifying the COI genes of two pest species using PCR technology.
