针刀髌周松解法对膝骨性关节炎兔线粒体凋亡通路的影响
Effect of Acupotomy Peripatellar Relaxation on Mitochondrial Apoptosis Pathway in Rabbits with Knee Osteoarthritis

Abstract
Objective:To explore the effect of patellar periarticular release under the guidance of the theory of muscular region on the mitochondrial apoptotic pathway in rabbits with knee osteoarthritis(KOA).Methods:Using the random number table method,18 healthy male New Zealand rabbits aged 6 months were randomly divided into blank group,model group and acupotomy group with 6 rabbits in each group.According to the modified Videman method,the left hind limbs of the rabbits in the model and acupotomy groups were immobilized for 6 weeks to prepare the rabbit KOA models.The total score of Lequesne MG behavioral assessment was≥4 points which indicted KOA models were established successfully.The acupotomy group selected the attachment points of the medial and lateral femoral muscles,the medial and lateral patellar support bands,the lateral patellar ligament on the patella,and the attach-ment point of the gastrocnemius tendon to the tibial medial condyle for acupotomy release,once a week for 4 times.The other two groups were raised normally and only underwent the same grasping and fixation without treatment.One week after the intervention,knee joint cartilage tissue samples were taken from the rabbits in all groups.Using toluidine blue staining,the morphological changes of the cartilage tissue were observed.Western blot and RT-qPCR were used to detect the relative protein expression levels and mRNA transcriptional levels of key apoptotic indicators,such as tumor protein P53(P53),B-cell lymphoma-2 protein(Bcl-2)related X pro-tein(Bax),cytochrome C(Cyt C),caspase-3,caspase-9,and anti-apoptotic key indicators such as Bcl-2 in the cartilage cells of the mi-tochondrial apoptotic pathway.Results:Compared with the blank group,the cartilage surface of the model group was rough,the ar-rangement of chondrocytes was loose and disordered,the number of chondrocytes decreased,there were obvious vacuoles,blue stain-ing decreased obviously,the tide line was blurred,the relative expression of P53,Bax,Cyt C,Caspase-3,Caspase-9 protein relative expressions and mRNA transcription levels increased(P<0.05),Bcl-2 protein relative expression and mRNA transcription level de-creased(P<0.05);Compared with the model group,the smoothness of cartilage surface improved,chondrocytes arranged more evenly and the number of which increased,blue staining decreased slightly,the fuzzy degree of tidal line improved,the relative expression of P53,Bax,Cyt C,Caspase-3,Caspase-9 protein relative expressions and mRNA transcription levels decreased(P<0.05),Bcl-2 protein relative expressions and mRNA transcription level increased in the acupotomy group(P<0.05).Conclusion:The acupotomy peripatel-lar relaxation method can affect the mitochondrial apoptosis pathway,effectively reduce the expression of apoptotic genes like P53,Bax,Cyt C,Caspase-3,and Caspase-9 in KOA rabbits,and increase the expression of anti-apoptotic gene Bcl-2,and delaying carti-lage degeneration.关键词
