ERK1/2信号通路在新生小鼠持续性肺动脉高压中发病的作用研究
The role of ERK1/2 signaling pathway in the pathogenesis of persistent pulmonary hypertension in neonatal mice

Abstract
Objective:To study the role of ERK1/2 signaling pathway in the pathogenesis of persistent pulmonary hyperten-sion PPHN in neonatal mice.Methods:A total of 80 neonatal mice were placed in a hypoxic box and randomly divided into the control group,PPHN group,DMSO group and inhibitor group.Right ventricular systolic pressure(RVSP)was measured by right ventricular puncture through the intercostal space,and the morphology of pulmonary blood vessels was observed by H&E staining,and pulmonary vascular remodeling indexes,such as the ratio of pulmonary vascular wall area to the total area of the tube(WA%)and the ratio of pulmonary arterial wall thickness to the outer diameter of the blood vessel(WT%)were calculated.Ex-pression level of proliferating cell nuclear antigen(PCNA)protein was detected by immunohistochemistry.The expression level of PCNA,ERK1/2 and p-ERK1/2 proteins in lung tissue was detected by Western blot.Results:The PPHN group exhibited ele-vated RVSP,increased right ventricular hypertrophy,increased alveolar destruction,significant thickening of pulmonary artery smooth muscle,and significant increased PCNA and p-ERK1/2 expression compared to the control group.In contrast,PPHN mice with the addition of the ERK1/2 inhibitor U0126 resulted in a reduction in PVSP,an improvement in right ventricular hyper-trophy,a reduction in alveolar destruction,and a significant decrease in pulmonary artery smooth muscle thickening.These effects were accompanied by a notable reduction of PCNA and p-ERK1/2 expression.Conclusion:The ERK1/2 signaling pathway is in-volved in the development of persistent pulmonary hypertension in neonatal mice.关键词
