基于组织病理学及子宫菌群探讨参芪扶正汤对慢性子宫内膜炎小鼠的影响
Effects of Shenqi Fuzheng Decoction on chronic endometritis in mice based on histopathology and uterine flora

Abstract
Objective:To investigate the effects and mechanisms of Shenqi Fuzheng Decoction(SQFZ)on chronic endometri-tis(CE)in mice.Methods:A total of 24 female SPF-grade BALB/c mice were randomly divided into 4 groups:the normal group,the model group,the SQFZ group,and the doxycycline group.Except for the normal group,CE mouse models were established in the other groups using the modified"intrauterine LPS infusion"method.The SQFZ group was administered a SQFZ solution(1 580 mg/mL),and the doxycycline group received a doxycycline solution(3 mg/mL).The normal and model groups were given an equivalent volume of distilled water by gavage once daily for 5 consecutive days.Mice were eathanized by cervical dislocation 24 h after the last administration.Histopathological changes were observed by H&E staining,CD138 expression was analyzed by immu-nohistochemistry,and 16S rRNA high-throughput sequencing was used to compare uterine microbiota communities among the groups.Results:H&E staining results showed that,compared to the normal group,the model group exhibited inflammatory changes,including plasma cell infiltration and stromal edema in the endometrium.In comparison with the model group,both the SQFZ and doxycycline groups had reduced inflammatory cell infiltration and alleviated inflammatory changes.Immunohistochemi-cal analysis of CD138-positive cells revealed that the expression rate of positive cells in the model group was significantly higher than in the normal group(P<0.05),while the positive expression rate in the SQFZ group was significantly lower than that in the model group(P<0.05).16S rRNA sequencing indicated no significant differences in species richness and diversity(α and β diver-sity)among the groups.However,species composition and differential analysis showed that,compared to the normal group,the abundance of Bacteroidetes was significantly reduced in both the model and doxycycline groups(P<0.05).Compared to the mod-el group,the SQFZ group showed an increase in Bacteroidetes abundance,approaching levels observed in the normal group.Con-clusion:SQFZ helps to improve the inflammatory condition of the endometrium in mice and regulates the uterine microbial ecosys-tem by increasing the abundance of Bacteroidetes,contributing to the restoration of microbial balance.关键词
