基于网络药理学和实验验证探讨藤茶-青钱柳对预防急性酒精中毒的作用机制
Mechanism of Ampelopsis grossedentata and Cyclocarya paliurus in preventing acute alcoholism based on network pharmacology

Abstract
Objective:To determine the potential mechanisms of action for Ampelopsis grossedentata and Cyclocarya paliurus in preventing acute alcohol poisoning(AAI)based on network pharmacology.Methods:Network pharmacology was utilized to predict potential targets and signaling pathways associated with the treatment of AAI using Ampelopsis grossedentata and Cyclo-carya paliurus.An AAI mouse model was established to validate the efficacy of Ampelopsis grossedentata and Cyclocarya paliurus in alleviating intoxication and protecting the liver in mice with AAI.Results:A total of 6 active components of Ampelopsis grossed-entata and Cyclocarya paliurus were identified,with core targets including AKT1,IL-6,EGFR,BCL-2,and CASP3,etc.The signaling pathways related to the treatment of AAI by Ampelopsis grossedentata and Cyclocarya paliurus were mainly concentrat-ed in cancer-related pathways,the PI3K/AKT signaling pathway,as well as pathways involved in toxoplasmosis.Animal experi-ments showed that compared to the model group,the latency period of intoxication was significantly prolonged and sleep time was significantly reduced in each treatment group(P<0.001).Compared to the control group,ALT,AST,and TG levels were sig-nificantly increased in the model group,along with elevated ADH and MDA levels in liver tissue,while the content of GSH and SOD was significantly decreased(P<0.001).Compared to the model group,serum ALT,AST,and TG levels were significant-ly decreased in the groups treated with Ampelopsis grossedentata and Cyclocarya paliurus,with significant increases in ADH,GSH,and SOD levels and a significant decrease in MDA content in liver tissue(P<0.001).Conclusion:Combined use of Am-pelopsis grossedentata and Cyclocarya paliurus exhibits excellent effects in preventing intoxication,accelerating alcohol metabo-lism,and protecting the liver,potentially through the activation of the PI3K/AKT signaling pathway.关键词
