寿胎丸联合菟丝子总黄酮三阴交贴敷对肾虚流产大鼠母胎免疫平衡因子的影响
Effects of of Shoutai Pill(寿胎丸)Combined withSanyinjiao(SP6)Application with Total Flavonoids of Tusizi(Cuscutae Semen)on Maternal Fetal Immune Balance Factors in Aborted Rats with Kidney Deficiency

Abstract
Objective To explore the effect of Shoutai Pill(寿胎丸)combined with Sanyinjiao(SP6)application with total fla-vonoids of Tusizi(Cuscutae Semen)on the immune balance factors in rats with kidney deficiency induced abortion.Methods Ex-cept for the normal control group,all the other groups were orally administered with hydroxyurea and mifepristone to establish a fe-male pregnant rat model of kidney deficiency induced abortion.Ten rats in the normal control group were not given a model,while 10 rats in the model group were given an equal amount of physiological saline by gavage daily and applied with starch paste at Sa-nyinjiao(SP6)for a total of 14 days;10 mice in the Shoutai Pill group(experiment group A)were given Shoutai Pill suspension by gavage daily during modeling,and starch paste was applied at Sanyinjiao(SP6);10 mice in the Sanyinjiao(SP6)application group(experimental group B)were given an equal amount of physiological saline by gavage every day while modeling,and the total fla-vonoid of Tusizi(Cuscutae Semen)was applied at Sanyinjiao(SP6);10 mice in the experimental group C were given Shoutai Pill suspension by gavage daily while modeling,and the total flavonoid of Tusizi(Cuscutae Semen)was applied to Sanyinjiao(SP6).After the experiment,the eye vein blood of the anesthetized rats were collected,and the serum was separated to detect the levels of prolactin(PRL)and progesterone(P).The uterine and decidual tissues were separated,and the expression of progesterone receptor(PR)in the decidual tissues were detected by using immunohistochemistry method.Immunohistochemical staining was used to de-tect the expressions of Fas/FasL,PCNA,HB-EGF and PR in the placenta and decidua.RT-PCR was used to detect the expres-sion of Th1/Th2 mR-NA in the decidua.Result:Compared with those of the control group,the miscarriage rate in the model group was significantly increased,and the total number of fetuses and average number of fetuses were significantly reduced(P<0.05).Compared with that of the model group,the application of Shoutai Pill and Sanyinjiao(SP6)application with total fla-vonoids of Tusizi(Cuscutae Semen)alone or in combination can reduce the miscarriage rate,increase the pregnancy rate and im-prove the fetal survival rate and embryo number(P<0.05).Compared with experimental group A and B,experimental group C had the lowest miscarriage rate,the highest pregnancy rate,fetal survival rate and embryo number(P<0.05).Compared with those in the normal group,the serum levels of PRL and P in the model group were significantly reduced(P<0.05).Compared with those in the model group,the levels of PRL and P in the serum of rats in experiment group A,B and C were significantly in-creased(P<0.05).Compared with the experimental group A and B,experimental group C had higher levels of PRL and P in the serum(P<0.05).Compared with that of the normal group,the PR expression level in the decidua of the model group was signifi-cantly reduced(P<0.05).Compared with those in the model group,the expression levels of PR in the decidua of rats in experi-mental group A,B and C were significantly increased(P<0.05).Compared with that in experimental group A and B,the PR ex-pression level in the decidua of experimental group C was higher(P<0.05).Compared with those in the normal group,the ex-pression levels of Fas and FasL in the decidua and placenta of the model group were significantly increased,while the expression levels of PCNA and HB-EGF in the decidua and placenta were significantly reduced(P<0.05).Compared with those of the model group,the expression levels of Fas and FasL in the decidua and placenta of experimental group A,B and C were significant-ly reduced,while the expression levels of PCNA and HB-EGF in the decidua and placenta were significantly increased(P<0.05).Compared with those of the experimental group A and B,the expression levels of Fas and FasL in the decidua and placenta of the experimental group C were lower,while the expression levels of PCNA and HB-EGF in the decidua and placenta were higher(P<0.05).Compared with those of the normal group,the mRNA expression levels of TNF-α and IFN-γ in the decidua of the model group significantly increased,while the mRNA expression levels of IL-4 and IL-10 significantly decreased(P<0.05).Compared with those in the model group,the mRNA expression levels of TNF-α and IFN-γ in the decidua of rats in the experimental group A,B and C significantly decreased,while the mRNA expression levels of IL-4 and IL-10 significantly in-creased(P<0.05).Compared with those of the experimental group A and B,the mRNA expression levels of TNF-α and IFN-γ in the decidua of experimental group C were lower,while the mRNA expression levels of IL-4 and IL-10 were higher(P<0.05).Conclusion The combination of Shoutai Pill and Sanyinjiao(SP6)application with total flavonoids of Tusizi(Cuscutae Se-men)can increase the pregnancy rate and improve pregnancy outcomes in rats with miscarriage.Its therapeutic mechanism may be related to regulating the balance of Th1/Th2 cytokines in the body of miscarriage rats and regulating the immune response imbal-ance in pregnant rats.关键词
