Correlation between Lymphocyte Subset Counts and Radiosensitivity in Lung Cancer

Abstract
Objective:To examine the pre-and post-radio-therapy lymphocyte subset profiles and their longitudinal trends in patients with primary and secondary non-small cell lung cancer(NSCLC),and to assess the association of these profiles with radiosensitivity.Methods:A retrospective analy-sis was conducted on 122 eligible patients with primary or secondary NSCLC who were treated at Sichuan Cancer Hospital from July 1,2019,to July 31,2024.Based on the recent efficacy evaluation,patients who achieved a complete response or partial response were designated as the radiosensitive group,whereas those with stable or progressive disease were assigned to the radioresistant group.The Chi-square test was utilized to compare the baseline and clinicopathological characteristics be-tween the two groups.Differences in lymphocyte subset counts between the radiosensitive and radioresistant groups were ana-lyzed with the independent samples t-test or Mann-Whitney U test.Spearman's rank correlation was then used to determine the correlation between lymphocyte subset counts and radiosensitivity.Results:In primary lung cancer,the radiosensitive group demonstrated significantly elevated levels in several parameters compared to the radioresistant group:the pre-and post-RT CD4+/CD8+ratio(P=0.041 and P=0.013,respectively),CD16+56+comparative counts(P=0.020 and P=0.033,respectively),and the absolute counts of CD3+CD4+(P=0.045 and P=0.025,respectively)and CD16+56+(P=0.002 and P=0.033,respectively).In metastatic lung cancer,the radiosensitive group showed significantly higher values than the radioresistant group for the following pre-RT parameters:the CD4+/CD8+ratio(P=0.035),and the abso-lute counts of CD3+(P=0.014),CD3+CD4+(P=0.042),and CD3+CD8+(P=0.038).Spearman correlation anal-ysis showed that in primary lung cancer,the pre-RT and post-RT CD4+/CD8+ratio(r=0.269,P=0.040;r=0.420,P=0.009),CD16+56+comparative counts(r=0.301,P=0.021;r=0.335,P=0.040),CD16+56+absolute counts(r=0.424,P=0.001;r=0.351,P=0.031)were positively correlated with radiosensitivity,and post-RT CD3+CD4+absolute counts were positively correlated with radiosensitivity(r=0.375;P=0.020).In metastatic lung cancer,the pre-RT CD4+/CD8+ratio was positively correlated with radiosensitivity(r=0.324;P=0.010).Conclu-sion:In primary NSCLC,the pre-RT and after-RT CD4+/CD8+ratios,CD16+56+comparative counts,CD16+56+abso-lute counts,and post-RT CD3+CD4+absolute counts are positively correlated with radiosensitivity.In metastatic lung canc-er,the pre-RT CD4+/CD8+ratio is positively correlated with radiosensitivity.关键词
