Effects of Rhizobium HH103 Effector Protein NopM on Heilongjiang Soybean Nodulation and Nitrogen Fixation Based on Transcriptome Analysis

Abstract
In order to investigate the effect of type Ⅲ effector NopM in Sinobium ferri HH103 on nodule fixation of soybean and the interaction mechanism between rhizobium and soybean,9 kinds of soybeans from Heilongjiang Province were used as experimental soybean materials,and rhizobium HH103 and its knockout mutant ΔnopM were inoculated on soybean roots respectively.To study the effect of nopM deletion on soybean and explore the changes of the transcriptome of the symbiosis between Qinong 7 soybean and rhizobia.With no inoculated rhizobium as a blank control,it was found that HH103 and ΔnopM both promoted nodulation in soybeans to a certain extent,but the nodulation effect was different in different soybean varieties.It is worth noting that compared with HH103,ΔnopM increased the number of nodules,fresh nodule weight and nitrogenase activity of Qinong 7.Transcriptome sequencing showed that 11 479 differentially expressed genes were identified in the roots of Qinong 7 soybean inoculated with HH103 and ΔnopM,including 6 326 up-regulated genes and 5 153 down-regulated genes.The deletion of nopM not only promoted the up-regulated expression of nodule genes,legumoglobin genes and other nodule nitrogen-fixation genes,but also affected the biological processes such as signal transduction and synthesis of secondary metabolites during the interaction between rhizobium and soybean.In addition,the absence of nopM also leads to the enrichment of differentially expressed genes in flavonoid synthesis and ABC transporter pathway.The results of this study provide novel research insights and theoretical references for the subsequent in-depth exploration of the molecular mechanisms by which the effector NopM regulates nodulation in soybean cultivar Qinong 7.关键词
