Abstract

In an attempt to deepen the exploration of soybean varieties grown in Hebei Province for resource materials resistant to soybean mosaic disease,this study was conducted with 28 Handan series soybean cultivars bred by the Handan Academy of Agricultural Sciences,inoculated with Soybean Mosaic Virus(SMV)strains SC3,SC7,SC8,and N3 prevalent in major soybean producing areas of Hebei Province,and screened for high-quality SMV-resistant soybean germplasm resources by examining the pathogenic phenotypes and identifying the disease index.The resistance identification results showed that the overall resistance of the Handou variety in this study to the SC8,N3,and SC3 strains was weaker compared to the SC7 strain.Among them,Handou 15 had stronger resistance to all SMV strains,while Handou 17 had weaker resistance to all SMV strains.RT-qPCR was employed to analyse the expression of three SMV-related genes(GmPR1,GmWRKY164 and GmRD22)in different resistant varieties at transcriptional levels.It was found that the expression of the three genes in Handan 15,which is more resistant,was significantly higher than that of Handan 17,which is less resistant to SMV,which is in line with the results of phenotypic identification.The differences in resistance to SMV of Handou cultivars may be closely related to the expression levels of disease resistance-related genes,which provides an experimental basis for the extensive screening of SMV-resistant resources.
