摘要

Abstract

[Objective]To provide a theoretical basis for further exploring the function of ZF-HD gene in the growth and development of Fagopyrum tataricum,the ZF-HD gene was cloned and its expres-sion pattern was analyzed.[Method]The FtZF-HD1 gene from Tartary buckwheat was cloned using reverse transcription polymerase chain reaction(RT-PCR).Bioinformatics analysis and real-time quantitative PCR(qPCR)were subsequently performed on the gene.[Result]The FtZF-HD1 coding sequence was 606 bp in length and encoded 201 amino acids.There was a homeobox and a zinc finger domain at the C-terminus and N-terminus of FtZF-HD1 protein,respectively.Phylogenetic analysis showed that FtZF-HD1 protein belonged to ZF-HD subfamily protein containing zinc finger domain and was most closely related to ZFHD1 protein in Arabidopsis thaliana.The result of subcellular localiza-tion prediction showed that FtZF-HD1 protein was localized in the nucleus.The q-PCR fluorescence quantitative analysis showed that FtZF-HD1 was expressed in different tissues of Tartary buckwheat,with the highest relative expression in seeds,and its expression pattern in seeds at different develop-mental stages showed a rapid increase followed by a slow decrease and stabilization.[Conclusion]FtZF-HD1 was highly expressed during the middle and late stages of Tartary buckwheat seed develop-ment,suggesting that it play an important regulatory role in the development of Tartary buckwheat seeds.
