摘要

Abstract

[Objective]Enrich the mitochondrial genome sequence data of the Nymphalinae,analyze the structural characteristics of the mitochondrial genomes in this subfamily,and clarify the phyloge-netic relationships among various taxonomic levels within the Nymphalinae.[Method]The mitochon-drial genomes of eight Nymphalinae species were sequenced employing high-throughput sequencing technologies.Phylogenetic analyses were performed based on 13 protein-coding genes and two ribo-somal RNA genes from the mitochondrial genome,with Sephisa chandra(Moore,[1858])and Apa-tura irisas(Linnaeus,1758)as outgroups.Maximum likelihood and bayesian inference methods were employed to construct a phylogenetic tree encompassing 49 species across 20 genera within the Nym-phalinae.[Result]The mitochondrial genomes of the eight Nymphalinae species were approximately 15 000 bp,consistent with those of other Lepidoptera.They were circular molecules comprising 37 genes and one control region,with an A+T content ranging from 80.2%to 81.4%.Among these Nym-phalinae species,except for the cox1 which starts with CGA,all other protein-coding genes started with ATN;the three codons with the highest relative synonymous codon usage(RSCU)were UUA,GGA,CGA;While AGC was the lowest.Microsatellite structure were present in the control regions of all eight species.The phylogenetic trees constructed by both methods had identical structures,each having two main branches:one exclusively containing Nymphalini,and the other branch's phylogeny was((Kallimini+(Melitaeini+Junoniini))+Victoriniini).Nymphalis ladakensis Moore,1882 and Inachis io(Linnaeus,1758)formed a close clade;Melitaeini included Doleschallia,Melitaea and Euphydryas.Yoma,Salamis and Junonia were monophyletic groups,but Yoma and Salamis had closer affinitis;Melicta ambigua Ménétriés,1859 and Melitaea bellona Leech,[1892]formed a clade and were sisters to Melitaea cinxia(Linnaeus,1758).[Conclusion]Nymphalinae is divided into five tribes:Nymphalini,Kallimini,Melitaeini,Junoniini,and Victoriniini.Doleschallia should be moved from Kallimini to Melitaeini and Mellicta ambigua should be classified into Melitaea.
