Abstract

The natural microbial community domestication strategy within the context of CBP(consolidated bioprocessing technology)was utilized to establish a targeted strain selection system,aiming to isolate thermophilic anaerobic spore-forming bacteria capable of lignocellulose degradation and cellulosic ethanol production.Via morphological examination,physiological and biochemical profiling,and 16S rDNA sequencing,strain XK1-1 was identified as Caldibacillus thermoamylovoran.The results show that under anaerobic conditions at 55 ℃ and 180 r/min,employing a glucose and xylose(mass volume ratio 2∶1 g/L)mixed carbon source during continuous cultivation for 12 h,strain XK1-1 realizes a maximal ethanol yield of(1.03±0.046)g/L,demonstrating heightened metabolic responsiveness to glucose.Furthermore,within a medium exclusively utilizing Salix as the carbon source and undergoing continuous fermentation for 48 h,the cellulose degradation rate by the strain uphold at 35%±0.21%,the hemicellulose degradation rate is at 53%±0.85%,and the ethanol yield is(0.27±0.05)g/L(significance p>0.05).Hence,this investigation furnishes high-quality strains and theoretical underpinnings for the advancement of non-food biomass fuels.
