Screening of Reference Genes of Pseudotaxus chienii by Real-time Fluorescent Quantitative PCR

Abstract
Pseudotaxus chienii,a monotypic genus within the Taxaceae family endemic to China,is a rare and protected species.To identify suitable reference genes for gene expression analysis in different tissues of P.chienii,this study selected six candidate reference genes(B1-B6)based on existing transcriptome data.Their expression stability was evaluated across various tissues(root,stem,old leaf,and new leaf)using real-time fluorescent quantitative PCR(RT-qPCR)and analyzed with five methods:geNorm,NormFinder,BestKeeper,comparative △Ct method,and RefFinder.The results indicated that B3 was the most stably expressed gene across all tested tissues,whereas B2 was the least stable.Thus,B3 is recommended as the optimal reference gene for future functional gene expression studies in P.chienii.关键词
