Abstract

To clarify the main pathogen species and basic biological characteristics of white mold on Morchella,diseased fruitbodies were collected from the morels cultivated at the Institute of Agricultural Sciences,in Linxia Hui Autonomous Prefecture,Gansu Province of China in this study.Five cultured mycelia were successfully isolated and purified by the tissue isolation method.Both field and in vitro inoculations were performed for the determination of pathogenicity by using Koch's rule.The results showed that four strains(ZX-1,ZX-2,WBY-4 and WBY-5)were from pathogens.Construction of phylogenetic trees on the basis of rDNA ITS sequences combining with the morphological observation identified them as the Pseudodiploospora longispora.Analysis of the biological characteristics revealed that the optimal temperature was 25℃ and the adaptation range of pH value was comparatively wider for the growth of pathogens,although they grew well in the alkaline condition and showed a certain tolerance to the acidic condition.The best carbon source of the pathogen was lactose,and the best nitrogen source was beef paste according to the supply test of carbon and nitrogen sources.The above results thus could provide a theoretical basis available for the future control of white mold in morel cultivation.
