摘要

Abstract

To explore functions of heat shock transcription factors responsable to environmental stress and growth development of macrofungi,five proteins carrying the DNA binding domain of Hsf family were identified from Pleurotus ostreatus,as named as PoHsf1-PoHsf5,respectively.Bioinformatics analysis showed that all of the five PoHsfs belonged to unstable and hydrophilic proteins and were located in the cell nucleus.Detection of conserved motifs and phylogenetic analysis using the full protein sequences indicated that PoHsf1 and PoHsf4 were highly related to ScHsf1 while PoHsf2 was closely similar to ScSkn7 of Saccharomyces cerevisiae.Analysis of gene expression patters demonstrated that the activity of PoHsf1 was significantly downregulated during the two stages of primordial formation and fruiting body as well as under the high temperature stress but was significantly upregulated under the low temperature and hydrogen peroxide stress.The expression activity of both PoHsf2 and PoHsf3 was decreased during the primordial formation stage.Significant upregulation of exression activity was observed for PoHsf4 under the high temperature stress and for PoHsf5 during the primordial formation and fruiting body stages,respectively.The above study thus should provide a useful foundation for future understanding of the functional roles of heat shock transcription factors particularly on the stress responses in P.ostreatus.
