Exploring the causal relationship between immune cells and intervertebral disc degeneration based on Mendelian randomization analysis methods

Abstract
Objective To explore the causal relationship between immune cells and intervertebral disc degener-ation using a two-way two-sample Mendelian randomization(MR)analysis methods.Methods Genetic varia-tion was used by MR as an instrumental variable to examine the causal effects of exposure factors on out-comes.In this paper,MR-Egger,inverse variance weighted and weighted median were used for MR analysis,and sensitivity analysis was performed to evaluate the robustness of the results.Results After false discovery rate(FDR)correction,two immunophenotypes significantly associated with disc degeneration were identified in this study:Leukocyte Antigen DR(HLA DR)on CD14+CD16-monocyte(OR=0.955,95%CI:[0.938,0.973],PFDR=1.26×10-4),and HLA DR on CD14+monocyte(OR=0.954,95%CI:[0.936,0.972],PFDR=2.49×10-4).In the reverse analysis,there was a negative correlation between intervertebral disc degeneration and two kinds of immune cells:CD25 on unswitched memory B cell(OR=0.844,95%CI:[0.713,0.999],PFDR=0.048),and CD34 on Hematopoietic Stem Cell(OR=0.668,95%CI:[0.519,0.860],PFDR=0.018).During the whole analyzing process,no evidence of heterogeneity or pleiotropy was observed,indicating that the results of this paper were robust.Conclusion This study confirmed the potential causal relationship between immune cells and intervertebral disc degeneration through Mendelian randomization analysis,and further improved the mechanism of immune cell-mediated intervertebral disc degeneration,which is expected to provide theoretical support for future clinical and basic research.关键词
