Aptamer SELEX Technology and Its Applications in Biomedicine
摘要
Abstract
Aptamers are artificially synthesized oligonucleotide fragments screened from oligonucleotide libraries through systematic evolution of ligands by exponential enrichment(SELEX)technology,and they possess the char-acteristics of high affinity and high specificity binding to targets.During the screening process,one of the key steps is to isolate the oligonucleotide fragments with strong binding ability to the target from the library,with the aim of obtaining a high-quality functional aptamer sequence.Since its inception,the SELEX technology has undergone continuous in-novation and improvement,covering a wide range of new technologies based on various targets and platforms such as cells,capillary electrophoresis,magnetic beads,graphene oxide,and microfluidic chips,and has been successfully applied to aptamers screening.This paper reviews the concept of aptamers,the differences between aptamers and antibodies,the diverse methods of aptamer screening,and their practical applications in the biomedical field,with the aim of providing a reference basis for the subsequent screening of aptamers and the application research in related fields.关键词
