Metabolomics Study on the Treatment of Cholestatic Hepatitis with Digeda-8 Powder

Abstract
By analyzing the metabolomics of cholestatic hepatitis treated by Digeda-8 powder with Lomatogo-nium as the monarch drug,this paper provides a basis for elucidating the correlation theory between monarch drug and target organs in Mongolian compound preparations.The LC-MS/MS system quadrupole-electrostatic field or-bital trap Orbitrap mass spectrometer was used to perform non-targeted metabolomics analysis on rat serum samples of blank group,model group,Lomatogonium group,and Digeda-8 powder group.The serum differential metabolites of each group of rats were detected,and then the possible regulatory metabolic pathways were analyzed.The differ-ential metabolites of each group were analyzed by Venny 2.1.0-CSIC database,and the results showed that 19 meta-bolic pathways were obtained in the blank-model comparison group,26 metabolic pathways were obtained in the model-Digeda-8 powder comparison group,and 47 metabolic pathways were obtained in the model-Lomatogonium comparison group.Correlation analysis of three groups obtained 12 metabolic pathways,including glycine,serine and threonine metabolism,aminoacyl-tRNA biosynthesis,biosynthesis of unsaturated fatty acids,phenylalanine me-tabolism,nitrogen metabolism,glycerophospholipid metabolism,arginine and proline metabolism,fatty acid metabo-lism,fatty acid biosynthesis,fatty acid elongation in mitochondrial,cytochrome P450 metabolism of exogenous sub-stances,steroid hormone biosynthesis and so on.It was found that the Lomatogonium regulation pathway accounted for 46%in the metabolic pathway of Digeda-8 powder in the treatment of cholestatic hepatitis.Therefore,Digeda-8 powder may play a role in the treatment of cholestatic hepatitis through the 12 metabolic pathways regulated by Lo-matogonium.关键词
