神经肽FF对乳腺浸润性导管癌细胞增殖和侵袭迁移的影响
Effects of neuropeptide FF on proliferation,invasion,and migration of invasive ductal carcinoma cells

Abstract
Objective To investigate the effects of neuropeptide FF(NPFF)on the proliferation,invasion,and migration of invasive ductal carcinoma(IDC)cells and the associated mechanisms.Methods We measured the level of NPFF in IDC tissue by enzyme-linked immunosorbent assay;determined the expression of NPFF and NPFF receptor 2(NPFFR2)in IDC tissue by immu-nohistochemistry;measured NPFF levels in IDC cell lines(MCF10A,MCF10AT,MCF10DCIS,and MCF10CA1d)by Western blot;assessed the effects of NPFF on the proliferation,migration,and invasion of MCF10A cells by Cell Counting Kit-8 assay,scratch assay,and Transwell assay;and measured the expression of proteins related to the extracellular signal-regulated kinase(ERK)1/2 signaling pathway by Western blot.Results Compared with those in adjacent tissue,the expression levels of NPFF and NPFFR2 proteins in IDC tissue were significantly increased(t=12.55,11.64,both P<0.001).The expression of NPFF pro-tein in IDC tissue was significantly higher than that in ductal carcinoma in situ tissue(t=5.34,P<0.001).MCF10CA1d cells had significantly higher NPFF and NPFFR2 protein levels than MCF10A cells(F=9.92,6.64,both P<0.05).Compared with NPFF at 0 μmol/L,NPFF at 20 and 40 μmol/L significantly increased cell viability and migration,and NPFF at 80 μmol/L significantly increased cell invasion and p-ERK1/2 protein expression(t=4.72,5.84,F=3.56,all P<0.05);and cells treated with NPFF at 80 μmol/L plus BIBP3226 at 80 μmol/L exhibited significantly decreased invasion ability compared with the 80 μmol/L NPFF group(P<0.05).NPFFR2 knockout significantly reduced the activation of ERK1/2 induced by NPFF at 80 μmol/L(t=6.43,P<0.05).Conclusion NPFF is highly expressed in IDC tissue,which promotes the proliferation,migration,and invasion of IDC cells by activating the ERK1/2 signaling pathway.关键词
