Effects of intranasal administration of ferric ammonium citrate on the expression of glucocerebrosidase and lysosomal integral membrane protein-2 in the olfactory bulbs of mice

Abstract
Objective To investigate the effects of intranasal administration of ferric ammonium citrate(FAC)on the ex-pression of glucocerebrosidase(GCase)and lysosomal integral membrane protein 2(LIMP-2)in mouse olfactory bulbs.Methods Forty-eight male C57BL/6 mice were randomly divided into 6 groups with 8 mice in each group.Three groups were given FAC bi-lateral alternate nasal drops(200 mg/kg)once every other day for 1,3,or 6 weeks,respectively.The 3 control groups were given the same amount of normal saline over the same time periods.Protein levels of light chain ferritin,GCase,and LIMP-2 in olfactory bulbs were detected by Western blotting.Results Compared with the control groups at 1,3,and 6 weeks,the expression of light chain ferritin in the olfactory bulbs of mice in the experimental groups at the corresponding time points was significantly in-creased(t=6.289-27.270,P<0.001).There were no significant differences in GCase protein expression between nasal adminis-tration of FAC and nasal administration of saline at 1,3,and 6 weeks(P>0.05).Compared with nasal administration of normal saline for 1 week,the expression of LIMP-2 in olfactory bulbs of mice given nasal administration of FAC for 1 week was significant-ly increased(t=4.483,P<0.01),but there were no significant differences in the expression of LIMP-2 at 3 and 6 weeks(P>0.05).Conclusion There was significant iron deposition in the olfactory bulbs after nasal administration of FAC,but it was in-sufficient to induce significant dysfunction of GCase or LIMP-2.关键词
