摘要

Abstract

Objective To investigate the effect of enriched environment(EE)on the motor and cognitive functions of mice with middle cerebral artery occlusion/reperfusion(MCAO/R)and its underlying mechanism.Methods A mouse model of MCAO/R was constructed,and then 1 day later,the mice were fed in EE or standard environment for 14 days.The modified Neu-rological Severity Score(mNSS)and the rotarod test were used to evaluate the motor function of mice;the Y-maze test was used to evaluate the cognitive function of mice;Western blotting was used to measure the expression level of vesicular glutamate trans-porter 1(VGLUT1)in the cortex,striatum,and hippocampus of the ipsilateral side.Results EE significantly reduced the mNSS score of MCAO/R mice(F=103.900,q=9.551,P<0.001)and increased fall latency in the rotarod test,the percentage of time spent in the novel arm,and the percentage of entries into the novel arm in MCAO/R mice(F=10.110-31.910,q=4.271-7.769,P<0.05),and it also reduced the expression level of VGLUT1 in the cortex,striatum,and hippocampus of the ipsilateral hemisphere of MCAO/R mice(F=8.141-14.600,q=5.318-7.240,P<0.05).Conclusion EE can significantly improve the motor and cognitive disorders in mice with MCAO/R,possibly by downregulating the expression of VGLUT1.
