Effect of enriched environment on the motor and cognitive functions of mice with ischemic stroke

Abstract
Objective To investigate the effect of enriched environment(EE)on the motor and cognitive functions of mice with middle cerebral artery occlusion/reperfusion(MCAO/R)and its underlying mechanism.Methods A mouse model of MCAO/R was constructed,and then 1 day later,the mice were fed in EE or standard environment for 14 days.The modified Neu-rological Severity Score(mNSS)and the rotarod test were used to evaluate the motor function of mice;the Y-maze test was used to evaluate the cognitive function of mice;Western blotting was used to measure the expression level of vesicular glutamate trans-porter 1(VGLUT1)in the cortex,striatum,and hippocampus of the ipsilateral side.Results EE significantly reduced the mNSS score of MCAO/R mice(F=103.900,q=9.551,P<0.001)and increased fall latency in the rotarod test,the percentage of time spent in the novel arm,and the percentage of entries into the novel arm in MCAO/R mice(F=10.110-31.910,q=4.271-7.769,P<0.05),and it also reduced the expression level of VGLUT1 in the cortex,striatum,and hippocampus of the ipsilateral hemisphere of MCAO/R mice(F=8.141-14.600,q=5.318-7.240,P<0.05).Conclusion EE can significantly improve the motor and cognitive disorders in mice with MCAO/R,possibly by downregulating the expression of VGLUT1.关键词
