Abstract

Objective To investigate the effect of checkpoint kinase 2(CHK2)on neurological function in rats with vascular dementia(VD)and its mechanism by regulating poly(ADP-ribose)polymerase 1(PARP1).Methods A total of 46 adult male Sprague-Dawley rats were divided into negative control group with 13 rats,model group with 13 rats,CHK2 shRNA group with 10 rats,and PARP1 shRNA group with 10 rats.HE staining was used to observe the pathological changes of hippocampal tissue and neurons in rats;TUNEL assay was used to detect the apoptosis of hippocampal neurons in rats;the Morris water maze test was used to evaluate the changes in the number of platform crossings and escape latency;ELISA was used to measure the expres-sion of interleukin-1β,interleukin-6,and tumor necrosis factor-α in the hippocampal tissue of rats;colorimetry was used to mea-sure the expression of superoxide dismutase(SOD),reduced glutathione(GSH),and malondialdehyde(MDA)in hippocampal tis-sue;Western blotting was used to measure the expression of CHK2 and PARP1 in hippocampal tissue.Results There were sig-nificant differences in the mean values of each indicator between multiple groups(F=46.850-607.692,P<0.001).Compared with the model group,the CHK2 shRNA group and the PARP1 shRNA group had improvement in the damaged hippocampal tis-sue structure,a significant reduction in the number of apoptotic hippocampal neurons(P<0.05),a significant increase in the num-ber of platform crossings,and a significant reduction in escape latency(P<0.05),as well as significant reductions in the expres-sion of inflammatory factors and MDA and significant increases in the expression of SOD and GSH in hippocampal tissue(P<0.05).The CHK2 shRNA group had a significant reduction in the expression of PARP1 in hippocampal tissue(P<0.05).Con-clusion Inhibiting the expression of CHK2 in VD rats can improve their cognitive function,possibly by interfering with CHK2 to reduce the expression of PARP1.
