解毒活血汤对CKD大鼠肾脏保护作用及对TGF-β1、α-SMA蛋白表达的影响
Protective effect of Jiedu Huoxue decoction on the kidney of CKD rats and impact on TGF-β1 and α-SMA protein expression

Abstract
Objective:To investigate the effects of Jiedu Huoxue decoction on renal function,Transforming Growth Factor-β1(TGF-β1),and α-Smooth Muscle Actin(α-SMA)in rats with chronic kidney disease(CKD).Methods:Seventy-two 7-week-old male SD rats of SPF grade were randomly divided into six groups(n=12 per group):Control group,Model group,High-dose Jiedu Huoxue decoction group(3.16 g/kg),Medium-dose Jiedu Huoxue decoction group(1.58 g/kg),Low-dose Jiedu Huoxue decoction group(0.79 g/kg),and Positive control group(Niaoduqing granule,1.81 g/kg).A CKD rat model was established using the 5/6 nephrectomy method.Drug administration via intragastric gavage began 8 weeks after modeling and lasted for 12 weeks.Blood was collected from all groups at the end of the 4th week of gavage for renal function parameter testing.After 12 weeks,TGF-β1 andα-SMA protein expression levels were detected using Western blot.Data were processed using SPSS 26.0 statistical software.Results:Compared with the Control group,serum calcium,phosphorus,urea nitrogen,creatinine,and TGF-β1 and α-SMA protein expression levels showed significant differences in all other groups(P<0.05 or P<0.01).Compared with the Model group,all Jiedu Huoxue decoction treatment groups exhibited significantly lower serum phosphorus,urea nitrogen,and creatinine(P<0.05),significantly higher serum calcium(P<0.05),and significantly reduced TGF-β1 andα-SMA protein expression(P<0.05 or P<0.01).Compared with the Low-dose group,the Medium-dose and High-dose groups showed significantly lower TGF-β1 andα-SMA protein expression(P<0.05).Compared to the High-dose group,the Low-dose and Medium-dose groups showed significantly higher TGF-β1 andα-SMA pro-tein expression(P<0.05).Conclusion:The therapeutic effect of Jiedu Huoxue decoction was accompanied by down-regulation of TGF-β1 andα-SMA protein expression,suggesting its action may be related to the suppression of the fi-brotic pathway.关键词
