朱顶红属植物不同组织部位基因组DNA提取效果比较分析
Comparative analysis of genomic DNA extraction efficiency from different tissue parts of Hippeastrum plants

Abstract
To investigate the DNA extraction efficiency from different tissue parts of Hippeastrum plants,genomic DNA was extracted and compared from 4 different parts(flower buds,sepals,inner leaves,and outer leaves)of 4 Hippeastrum varieties('Tika Flamenco','Tika Gloriosa','Tika Disco',and'Tika Fantasia')using the CTAB method.The DNA testing results showed that the extracted DNA concentration from the tender flower buds of Hippeastrum was 1 164.28-1 310.73 ng/μL,with an OD260/280 ratio of 1.906-1.937.The DNA extracted from the inner leaves had a concentration of 1 460.98-2 476.58 ng/μL and an OD260/280 value of 1.863-1.964,along with an OD260/230 ratio of 1.930-2.050.Although the DNA concentration extracted from the inner leaves and outer leaves met expectations,the purity was relatively low.The concentration of DNA extraction from different parts,from high to low,was as follows:inner leaves,flower buds,outer leaves,sepals;the purity from high to low was:inner leaves,flower buds,sepals,outer leaves.The bands amplified by PCR primers were clear and bright,and the inner and outer leaves performed better than the flower buds and sepals.This study comprehensively evaluated the quality,concentration,and PCR amplification products of DNA,and determined that the inner leaves were the optimal DNA extraction site for Hippeastrum.This laid the foundation for subsequent molecular marker assisted selection breeding,stress resistance breeding,variety identification,and other work based on DNA sequencing,which was of great significance for promoting research on Hippeastrum plants.关键词
