Analysis of the mechanism of Fengshi Gutong Capsule on immune inflammation in osteoarthritis based on data mining technology and network pharmacology

Abstract
Objective:To investigate the clinical efficacy and mechanism of action of Fengshi Gutong Capsule(FSGTC)in treating osteoarthritis(OA).Methods:Data mining was used to explore the clinical efficacy of FSGTC in treat-ing OA.Network pharmacology technology was used to construct the"active ingredient-target-disease"interaction network map,analyze the interaction between related proteins,and perform enrichment analysis of the biological processes of gene pathways.Additionally,molecular docking was conducted using AutodockTools 1.5.7 and PyMOL.Results:(1)Compared to the non-FSGTC group,the FSGTC group showed significantly lower levels of hypersensitive C-reactive protein(hs-CRP),immunoglobulin M(IgM)and erythrocyte sedimentation rate(ESR),with statistically significant differences(P<0.01).According to association rule analysis,FSGTC was closely associated with the reduction of hs-CRP,complement C3(C3),complement C4(C4),immunoglobulin A(IgA),immunoglobulin G(IgG)and ESR levels.Similar results were vali-dated using a random walk model.(2)A total of 109 active ingredients were screened,and the drug acted on 145 disease targets.Key targets such as JUN,TNF,MAPK14,MAPK1,MAPK3,TP53,Akt1,RELA,STAT3 and FOS interacted to form a core target network for the therapeutic action of the drug.Additionally,pathways such as AGE-RAGE,MAPK and HIF-1 were found to influence the inflammatory response in OA.(3)Molecular docking results indicated that the binding energies of kaempferol,quercetin and formononetin to the JUN receptor were-7.50,-7.80 and-7.10 kcal/mol,respectively.Conclusion:FSGTC can effectively ameliorate immune inflammation in OA,which may be associated with the combined ac-tion of active ingredients such as kaempferol,quercetin and formononetin.关键词
