摘要

Abstract

Objective:To investigate the reproductive toxicity and underlying molecular mechanisms of Tripterygium glycosides(TG)in female rats using proteomic analysis.Methods:Sixteen specific-pathogen-free(SPF)female Sprague Dawley(SD)rats aged 8 weeks were randomly divided into a TG group and a control group(n=8 per group)according to a random number table.The TG group received TG by gavage at 50 mg/(kg·d),while the control group received normal saline.The estrous cycle was mo-nitored during the gavage period.After continuous administration for 14 days,body weight and organ indices were recorded.Serum hormone levels were measured by enzyme-linked immunosorbent assay(ELISA),and histopathological changes in the ovary and u-terus were observed by hematoxylin-eosin(HE)staining.Label-free quantitative proteomic analysis was performed on ovarian tis-sues from both groups,and key differentially expressed proteins(DEPs)enriched in significant pathways were validated by Western blot.Results:TG significantly prolonged the estrous cycle(P<0.000 1),increased serum levels of luteinizing hormone(LH)(P<0.000 1)and follicle-stimulating hormone(FSH)(P<0.000 1),while reducing estradiol(E2)(P<0.01),progesterone(P)(P<0.000 1),and testosterone(T)(P<0.01)levels.Ovarian atrophy and a marked increase in the rate of atretic follicles(P<0.000 1)were observed.Proteomic analysis identified 203 DEPs,mainly involved in metabolic pathways,insulin signaling,and the P53 signaling pathway.Western blot validation showed that TG upregulated tumor protein P53(P53),cytochrome C,mitochondrial apoptosis-related gene Bcl2-associated X protein(Bax),caspase-3(Caspase-3),and apoptosis-inducing factor mitochondria-associ-ated 2(Aifm2),while downregulating B-cell lymphoma 2(Bcl-2).Conclusion:TG induces reproductive toxicity in female rats,and its mechanism may be associated with modulation of the P53 signaling pathway.
