Effect of Genistein on Non-small Cell Lung Cancer Based on Bioinformatics Analysis

Abstract
Objective:To investigate the effect of genistein on non-small cell lung cancer(NSCLC)A549 cells and its potential therapeutic targets through bioinformatics analysis and cell biology experiments.Methods:A549 cells were divided into a control group and a genistein treatment group(100 μmol/L).After 48 hours of treatment,RNA was extracted using an RNA extraction kit and subjected to transcriptome sequencing.Differentially expressed genes were analyzed using heatmaps and volcano plots on the Sangerbox website.Gene Ontology(GO)enrichment analysis was performed using the Metascape website.Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis was conducted using OmicShare.Drug-disease intersecting targets were uploaded to Cytoscape v3.7.2 for protein-protein interaction(PPI)analysis.Flow cytometry was used to validate the effect of genistein on the cell cycle of A549 cells.Results:Compared with the control group,genistein treatment altered mRNA expression,with significant differences between the two groups.A total of 4 630 differentially expressed genes were identified,including 2 853 upregulated genes and 1 777 downregulated genes.KEGG enrichment analysis indicated that genistein affected the cell cycle.Flow cytometry re-sults demonstrated that genistein induced G2/M phase arrest in A549 cells and simultaneously affected mitotic progression.Conclu-sion:Genistein may inhibit NSCLC progression by influencing the A549 cell cycle,DNA replication,and p53-related signaling path-ways.关键词
