参天益智咀嚼片的制备及其防治阿尔茨海默病作用评价
Preparation of Shentian Yizhi Chewable Tablets and Evaluation of Efficacy in Treatment of Alzheimer's Disease

Abstract
Objective:To optimize the preparation process of Shentian Yizhi Chewable Tablets and to preliminarily explore their therapeutic effects and mechanisms in the treatment of Alzheimer's disease(AD).Methods:The inclusion rate of volatile oil,yield of inclusion complex,extract yield,and extraction efficiency of active ingredients were used as evaluation indices.The inclusion process of volatile oil,water decoction process,and tablet formulation process were optimized using the Box-Behnken response sur-face methodology and orthogonal experimental design.An AD zebrafish model was established to evaluate the efficacy of Shentian Yizhi Chewable Tablets in restoring motor dysfunction,improving response ability,and inhibiting acetylcholinesterase(AChE)activ-ity.Results:The optimal inclusion process was achieved with a host-guest ratio([3-cyclodextrin:volatile oil)of 8:1,using a saturat-ed aqueous solution method at 50 ℃ for 3 h.The optimal extraction process involved two decoctions,each with 5 times the volume of water,boiled for 90 min.The optimal formulation ratio of Shentian Yizhi extract to microcrystalline cellulose,mannitol,and citric acid was 20:5:10:0.5.The tablets were prepared by wet granulation and compression,each weighing 1.5 g with a hardness of 50 to 70 N.Zebrafish experiments showed that,compared with the model group,the high-dose Shentian Yizhi Chewable Tablets group exhibited a significantly increased total swimming distance,a significantly higher light-dark velocity difference per minute,and a significantly reduced acetylcholinesterase fluorescence intensity.Conclusion:The preparation process of Shentian Yizhi Chewable Tablets is stable and feasible.This formulation shows potential therapeutic effects against Alzheimer's disease and holds promise for development as a functional food for AD prevention and treatment.关键词
