Mechanisms of Immune-enhancing Effect of Zhenyuan Capsules on Rats with Spleen and Kidney Deficiency Based on Syndrome-efficacy Correlation

Abstract
Objective:To investigate the mechanisms by which Zhenyuan Capsules enhance immunity through strengthening the spleen and kidney in rats with spleen-kidney deficiency induced by hydrocortisone combined with senna leaf based on correlation between syndrome and efficacy.Methods:Male Sprague Dawley rats were randomly divided into five groups according to a random number table:normal group,model group,positive drug group,low-dose Zhenyuan Capsules group,and high-dose Zhenyuan Cap-sules group.The immunodeficiency and spleen-kidney deficiency model was established by intramuscular injection of hydrocortisone combined with gradually increasing doses of senna leaf.Enzyme-linked immunosorbent assay(ELISA)was used to determine the serum levels of immunoglobulin A(IgA),immunoglobulin M(IgM),immunoglobulin G(IgG),complement components C3 and C4,interleukin-6(IL-6),interleukin-10(IL-10),and tumor necrosis factor-α(TNF-α).Immunofluorescence staining was employed to observe the expression and distribution of tight junction proteins,including zonula occludens-1(ZO-1),claudin-1,and occludin in colon tissues.Flow cytometry was used to analyze lymphocyte subsets in peripheral blood.Results:Compared with the model group,the ZO-1 fluorescence intensity was significantly increased in all medication groups(P＜0.05),while claudin-1 and occludin fluo-rescence intensities were markedly elevated in the high-dose Zhenyuan Capsules group(P＜0.05).The proportions of CD3+and CD4+T cells,as well as the CD4+/CD8+ratio,were significantly increased in all treatment groups(P＜0.05),whereas the pro-portion of CD8+T cells was significantly decreased(P＜0.05).Levels of TNF-α and IL-6 were markedly reduced in all treatment groups(P＜0.001),while IL-10 levels were significantly increased in the low-and high-dose Zhenyuan Capsules groups(P＜0.05).IgA levels were significantly elevated in the positive drug and high-dose Zhenyuan Capsules groups(P＜0.01);IgG and C3 levels were significantly increased in all treatment groups(P＜0.05);and IgM levels were significantly higher in the low-dose Zhenyuan Capsules and positive drug groups(P＜0.05).Conclusion:Zhenyuan Capsules exert spleen-and kidney-strengthening effects by enhancing immunity through elevating immunoglobulin and complement levels.The underlying mechanisms may be asso-ciated with repairing intestinal mechanical barrier damage,regulating lymphocyte subset balance,and modulating inflammatory me-diators.关键词
