基于河马信号通路研究骨病回生丸对激素性股骨头坏死的作用机制
Mechanism of Gubing Huisheng Pills in the Treatment of Steroid-induced Osteonecrosis of the Femoral Head Based on Hippo Signaling Pathway

Abstract
Objective:To investigate the mechanism of action of Cubing Huisheng Pills on steroid-induced osteonecrosis of the femoral head(SONFH)in rats via the Hippo signaling pathway.Methods:A rat model of SONFH was established by tail vein in-jection of lipopolysaccharide(LPS)combined with intraperitoneal injection of methylprednisolone.Seventy-two Sprague-Dawley(SD)rats were randomly assigned to the blank control group,model group,Xianling Gubao Capsules group,and low-,medium-,and high-dose Cubing Huisheng Pills groups.One week after model establishment,treatments were administered.At week 8,femoral bone structural strength was measured.Femur sections were stained with hematoxylin-eosin(HE).Micro-computed tomography(Mi-cro-CT)was used to assess bone microarchitecture.Serum alkaline phosphatase(ALP),osteocalcin(BGP),and calcium(Ca)lev-els were measured.Expression of Yes-associated protein(YAP)and transcriptional coactivator with PDZ-binding motif(TAZ)was assessed by Western blot and immunohistochemistry.Results:Compared with the model group,body weight of rats in the Xianling Gubao and Cubing Huisheng Pills groups showed an increasing trend.The model group exhibited reduced femoral bone strength,thinner trabeculae,increased incidence of empty lacunae,decreased bone volume fraction(BV/TV),trabecular thickness(Tb.Th),and trabecular number(Tb.N),increased trabecular separation(Tb.Sp),decreased serum ALP,BGP,and Ca levels,and reduced YAP and TAZ expression in femoral tissue.Compared with the model group,the Xianling Gubao groups and Cubing Huisheng Pills groups showed increased femoral structural strength,increased trabecular number,decreased incidence of empty lacunae and Tb.Sp,increased BV/TV,Tb.Th,Tb.N,serum ALP,BGP,Ca levels,and elevated YAP and TAZ expression in femoral tissue.Conclu-sion:Cubing Huisheng Pills improve femoral head structure and bone metabolism in SONFH in rats,potentially through modulation of the Hippo signaling pathway.关键词
