Effects of lncRNA TRHDE-AS1 regulating miR-340-5p/B3 GNT5 axis on proliferation,migration and invasion of liver cancer cells

Abstract
Objective:To investigate the effects of long non-coding thyrotropin releasing hormone degrading en-zyme antisense RNA1(lncRNA TRHDE-AS1)on migration,proliferation and invasion of liver cancer cells by regu-lating miR-340-5p/beta-1,3-N-acetylglucosaminyltransferase 5(B3 GNT5).Methods:The expression of lncRNA TRHDE-AS1,miR-340-5p and B3 GNT5 protein in liver cancer tissues,adjacent tissues and various liver cancer cells(HuH-7,HCCLM3,HepG2)was detected by RT-qPCR and Western blot.HepG2 cells were separated into blank group,si-NC group,si-lncRNA TRHDE-AS1 group,miR-NC group,miR-340-5p group,si-lncRNA TRHDE-AS1+anti miR-NC group,and si-lncRNA TRHDE-AS1+anti miR-340-5p group.RT-qPCR was applied to detect the ex-pression of lncRNA TRHDE-AS1,miR-340-5p,PCNA,MMP-9,and MMP-2 mRNA.Western blot was applied to de-tect the expression of B3 GNT5 protein.Clone formation experiment,CCK-8 experiment,scratch healing experiment,and Transwell experiment were applied to detect cell proliferation,migration,and invasion,respectively.The targeting relationship between lncRNA TRHDE-AS1 and miR-340-5p,as well as between miR-340-5p and B3 GNT5 was veri-fied by using the dual luciferase gene reporter assay.Results:Silencing lncRNA TRHDE-AS1 or overexpressing miR-340-5p was able to inhibit HepG2 cell proliferation,migration,and invasion.Down-regulation of miR-340-5p weak-ened the inhibitory effect of silenced lncRNA TRHDE-AS1 on malignant behavior of HepG2 cells.LncRNA TRHDE-AS1 targeted the regulation of miR-340-5p/B3 GNT5 axis.Conclusion:Silencing lncRNA TRHDE-AS1 can inhibit the proliferation,migration and invasion of HepG2 cells.Its molecular mechanism may be related to the upreg-ulation of miR-340-5p and the inhibition of B3 GNT5 expression.关键词
