丙泊酚调节MAPK/ERK信号通路对IL-1β诱导的软骨细胞损伤的改善作用实验研究

Abstract
Objective:To explore the ameliorative effect of propofol(PRO)on IL-1β-induced chondrocyte injury by regulating the MAPK/ERK signaling pathway.Methods:Human chondrocytes C-28/I2 were cultured in vitro and randomly classified into Ctrl group,IL-1βgroup,low-dose PRO group(12.5 μmol/L PRO),medium-dose PRO group(25 μmol/L PRO),high-dose PRO group(50 μmol/L PRO),and high-dose PRO+Chicolic acid group(50μmol/L PRO+10μmol/L MAPK/ERK signaling pathway activator Chicolic acid).The control group was cul-tured routinely,while the other groups were induced with 10 ng/ml IL-1β.CCK-8 method was used to measure the proliferation of C-28/I2 cells.Flow cytometry was performed to measure apoptosis in C-28/I2 cells.The reagent kit was used to measure oxidative stress indicators in the supernatant.ELISA method was used to detect the inflammatory factors in the supernatant.Western blot was performed to detect the MAPK/ERK signaling pathway proteins in C-28/I2 cells.Results:Compared with the Ctrl group,the IL-1βgroup showed a decrease in C-28/I2 cell survival rate,GSH-Px in cell supernatant,and an increase in apoptosis rate,MDA,IL-6,TNF-α levels,p-p38 MAPK/p38 MAPK and p-ERK1/2/ERK1/2 proteins(all P<0.05).Compared with the IL-1βgroup,the low-,medium-and high-dose PRO groups showed an increase in C-28/I2 cell survival rate,GSH-Px in cell supernatant,and a decrease in apoptosis rate,MDA,IL-6,TNF-αlevels,p-p38 MAPK/p38 MAPK and p-ERK1/2/ERK1/2 proteins,and the changes were dose-dependent(all P<0.05).The high-dose PRO+Chicolic acid group showed a decrease in C-28/I2 cell survival rate,GSH-Px in cell supernatant,and an increase in apoptosis rate,MDA,IL-6,TNF-αlevels,p-p38 MAPK/p38 MAPK and p-ERK1/2/ERK1/2 proteins than the high-dose PRO group(all P<0.05).Conclusion:PRO can ameliorate IL-1β-induced chondrocyte injury by inhibiting MAPK/ERK signaling pathway.关键词
