Expression profiles of premelanosome protein,guanylate binding protein 2,and Ki-67 in skin cutaneous melanoma and their association with clinicopathological characteristics and prognosis

Abstract
Objective To investigate the expressions of premelanosome protein(PMEL),guanylate binding protein 2(GBP2),and Ki-67 in skin cutaneous melanoma(SKCM)and their relationship with the clinicopathological characteristics and prognosis of SKCM pa-tients.Methods The mRNA levels of PMEL,GBP2,and Ki-67 in SKCM tissues(n=461)and normal skin tissues(n=558)and the cor-responding prognosis data of the SKCM patients were analyzed via the Gene Expression Profiling Interactive Analysis(GEPIA)database.Using a customized tissue microarray(TMA)of 50 SKCM patients,the expressions of PMEL,GBP2,and Ki-67 in SKCM tissues were detected by immunohistochemical staining,and their relationship with the clinical features and prognosis of the patients was analyzed.Results The analysis of the GEPIA database showed that both PMEL and Ki-67 were highly expressed in the SKCM tissues(both P<0.05),while GBP2 was lowly expressed(P>0.05).In the TMA analysis,the follow-up time for all patients ranged from 9 to 59 months.At the end of the follow-up,22 patients died,with an overall survival rate of 56.0%and a 3-year overall survival rate of 64.0%.Comparison among melanoma pathological types in the TMA showed that patients with the lentigo maligna type had the worst prognosis,while those with the acral lentiginous type had the best prognosis,and the prognosis of patients the nodular type and superficial spreading type was com-parable.The expressions of both PMEL and Ki-67 were the highest in lentigo maligna tissues,while GBP2 expression was the highest in acral lentiginous tissues.In the TMA analysis,the expressions of PMEL and Ki-67 in the tissues of the deceased at the end of the follow-up were both higher than those in the tissues of the surviving patients,whereas the expression of GBP2 was lower(all P<0.05).The TMA anal-ysis showed that high PMEL expression,low GBP2 expression,distant metastasis,and American Joint Committee on Cancer(AJCC)stage were independent risk factors affecting the overall survival of SKCM patients(all P<0.05).Conclusions High expressions of PMEL and Ki-67 in postoperative SKCM patients suggest a poor prognosis,while high expression of GBP2 suggests a good prognosis.These proteins can serve as potential prognostic markers for SKCM patients.关键词
