补中益气汤调控ULK1/FUNDC1介导的线粒体自噬防治骨质疏松症的机制研究
Research on the Mechanism of Buzhong Yiqi Decoction(补中益气汤)in regulating ULK1/FUNDC1-mediated Mitophagy for the prevention and treatment of Osteoporosis

Abstract
Objective To investigate the effect of Buzhong Yiqi Decoction(补中益气汤,BZYQD)on ULK1/FUNDC1-mediated mitophagy in the musculoskeletal tissues(femur and gastrocnemius muscle)of osteoporosis(OP)model rats and to elucidate its underly‐ing mechanism in OP prevention and treatment.Methods SD rats were allocated into the blank control group,model group,BZYQD group,and Western medicine(WM)group.Except for the blank control group,rats in the other groups underwent tail suspension to establish the OP model.Drug administration commenced concurrently with model establishment.After eight weeks,abdominal aortic blood samples(for serum),left femurs,and left gastrocnemius muscles were collected from rats in each group.Serum levels of alkaline phosphatase(ALP)and tartrate-resistant acid phosphatase(TRACP)were measured by ELISA;Micro-computed tomography(Micro-CT)was performed to analyze the morphology of the left femoral distal metaphysis and to calculate bone mass and trabecular parameters.HE staining and Masson staining were employed to observe pathological morphological changes in the femur and gastrocnemius muscle.Immunohistochemistry(IHC)was performed to detect Beclin-1 protein expression in the femur and gastrocnemius muscle;Levels of ROS,T-SOD,and MDA in the femur and gastrocnemius muscle were determined using specific kits:ROS by ELISA,T-SOD by the WST-1 method,and MDA by the thiobarbituric acid(TBA)method.Western blot(WB)and real-time quantitative polymerase chain reaction(RT-qPCR)were utilized to assess the protein and mRNA expression levels,respectively,of key molecules in the ULK1/FUNDC1 pathway within the femur and gastrocnemius muscle.Results Compared with the blank control group,the model group exhib‐ited significant alterations in musculoskeletal parameters,oxidative stress markers(ROS,T-SOD,MDA),and the ULK1/FUNDC1 pathway.However,intervention with BZYQD ameliorated these alterations to varying degrees and also alleviated the pathological mor‐phological damage observed in the femur and gastrocnemius muscle.Conclusion BZYQD may exert preventive and therapeutic effects against OP,potentially through modulating ULK1/FUNDC1-mediated mitophagy and ameliorating oxidative stress in the femur and gas‐trocnemius muscle.关键词
