右归丸含药血清通过p38/NF-κB抑制TNF-α诱导软骨细胞凋亡的机制研究
Mechanism of Yougui Pill(右归丸)-containing serum in inhibiting TNF-α-induced chondro-cyte apoptosis through the p38 MAPK/NF-κB signaling pathway

Abstract
Objective To investigate the mechanism by which Yougui Pill(右归丸,YGP)regulates autophagy to inhibit apoptosis of chondrocytes induced by TNF-α through the p38 MAPK/NF-κB signaling pathway.Methods Primary rat chondrocytes were divided into the untreated group,model group(TNF-α-induced),YGP-containing serum(YGP serum)group,and the control group.Cell prolifera‐tion was assessed using the CCK-8 assay.Apoptosis was detected by TUNEL assay.Levels of IL-1β and IL-6 in the chondrocyte super‐natant were measured by ELISA.Immunofluorescence(IF)was used to determine the expression of Beclin1,microtubule-associated protein 1 light chain 3 beta(LC3B),B-cell lymphoma 2(Bcl-2),and Bcl-2-associated X protein(Bax).Western blot(WB)was per‐formed to detect the protein expression levels of phosphorylated p38 MAPK(p-p38 MAPK),phosphorylated p65(p-p65),Beclin1,the ratio of LC3B-Ⅱ to LC3B-I(LC3B-Ⅱ/LC3B-I),Bcl-2,Bax,and Caspase-3.Results Compared with the model group,both the YGP serum group and the control group exhibited:significantly reduced inhibition of chondrocyte proliferation(P<0.05);significantly decreased chondrocyte apoptosis rates(P<0.05);significantly lower levels of IL-1β and IL-6 in the supernatant(P<0.05);signifi‐cantly lower expression of Bax,alongside significantly higher expression of Bcl-2,Beclin1,and LC3B(P<0.05);significantly lower protein expression levels of p-p38 MAPK,p-p65,Bax,and Caspase-3(P<0.05);and a significantly higher LC3B-Ⅱ/LC3B-I ratio(P<0.05).No significant differences were observed in any of these indicators between the YGP serum group and the control group(P>0.05).Conclusion YGP-containing serum regulates autophagy in chondrocytes,inhibits chondrocyte apoptosis,alleviates the inflammatory response,and protects chondrocytes by inhibiting the p38 MAPK/NF-κB signaling pathway.关键词
